CLOSE LOOK AT GENES: GENETIC ENGINEERING
Genetic engineering is already bringing relief to some people and hope to others. Fred Kostaras, of Levittown, Pennsylvania, has diabetes. He injects himself every morning and evening with a genetically engineered form of insulin that is the exact molecular duplicate of human insulin. Tracy Moreno, 10, who was growing too slowly, gets regular injections of growth hormone, cloned in the laboratory from human genes. Now, says Dr. Selma Kaplan, of the Pediatric Endocrine Unit at the University of California, San Francisco School of Medicine, Tracy is not fated to go through life as a 4-foot, 10-inch adult.
And there is hope for Marion Bennett, of Cheraw, South Carolina, who was stricken with melanoma in 1973. Despite surgery, the deadly skin cancer spread throughout her body. She is counting on injections of genetically engineered interferon to pull her through. Interferon is part of the body’s natural defense system in combating illness.
To create insulin, human growth hormone, and interferon, genetic engineers have taken a page from the beer brewer’s manual. Brewers use yeast to make beer because the yeast cell has a gene that controls the manufacture of alcohol. In the same way, genetic engineers can use yeast or bacteria – two simple germs-to create “natural” drugs. To make insulin, for example, they first isolate the gene for insulin from human cells. They splice the insulin gene into the yeast or bacteria genes. Those fast-growing microbes then turn out the human chemical, in this case, insulin. And because bacteria and yeast are easy to grow and reproduce so quickly, they can turn out limitless supplies of once scarce human hormones or chemicals.
Genetic engineers most recently cloned anti-hemophilic factor, the substance that helps blood clot, which hemophiliacs lack. Other “natural” drugs that may soon be genetically engineered include the following:
Ђў Endorphin – a natural morphine-like compound that controls pain and mood
Ђў Interleukin – a protein that regulates the body’s immune system
Ђў Human serum albumin-another protein available from blood that is used by doctors to replace blood loss from surgery or injury
Ђў Streptokinase and urokinase – two enzymes that work within the blood system, dissolving blood clots in the body, particularly the heart, brain, and lungs. Both are available now
Ђў Tissue plasminogen activator – a protein that dissolves blood clots, working at the site of the clot without affecting the blood supply
Ђў Erythropoietin – a natural human protein that stimulates the production of red blood cells
